PEI precipitation usage strategy:
1. PEI precipitation was used at high salt concentration (1mol/L NaCl). This method will precipitate nucleic acids while leaving almost all proteins in the supernatant.
2. When the target proteins are neutral and alkaline proteins, precipitation with PEI under 0.1mol/L NaCl conditions can remove acidic proteins and nucleic acids. At the same time, the target protein remains in the supernatant.
3. When the target protein is a strongly acidic protein (E. Coli RNA polymerase), a、 First, precipitate all acidic proteins and nucleic acids using a low concentration salt solution (0.1mol/L NaCl); b、 Dissolve and remove slightly acidic solution c with a medium concentration salt solution (0.4mol/L): Dissolve nucleic acids and strongly acidic proteins with a high concentration salt solution (0.9mol/L NaCl); d: Ammonium sulfate precipitates strongly acidic proteins.
